The lens paper was being imaged to provide a quick proxy for assessing the overall microscope health. This was almost a subjective check: "If I can see lens paper at 15 mW then things can't be too bad". So the data were acquired with just one channel.
I think we now have a better plan: Image lens paper with all channels at some reasonably high laser power and calculate photons/pixel. Possibly repeat this at a few gains for sanity. The reported result would therefore be the variance/mean fit plus, if possible, a standard light source converted to photons at that same gain and pixel size.
